No.

When you generate an instance, VS.NET will just generate some instance
filled with dummy data that can validate against the schema. This has
nothing to do with any instances you might allready have on file.

If the schema is a flat file schema, though, you might want to set the
"Generate Output Instance Type" to "Native" to generate a flat file
instead of XML.
